The Huygens Deconvolution wizard guides you through the process of microscopy image deconvolution in several stages and offers extensive help. Deconvolution settings and parameters are automoatically determined by Huygens for each channel and they allow editing.
Deconvolution Express performs unsupervised deconvolution with settings that are automatically determined for each image, and allows you to adjust the Acuity parameter for each channel. Deconvolution wizard guides you through the process of microscopy image deconvolution in several stages with the exact same deconvolution settings. However, unlike Deconvolution Express, you can adjust all the parameters within the Wizard.
The Operation Window in Huygens Professional (under Tools menu) allows you to adjust all the deconvolution parameters for all channels and run the deconvolution on all channels instantly. You can also create a deconvolution template and use this in the first step of the Deconvolution Wizard or in the Workflow Processor. You can create this template in the last step of the Deconvolution Wizard or from scratch in the deconvolution template editor tool that can be found via the "Edit" top menu of the main window.
In the last stage of the Deconvolution Wizard there is an option to store the used parameters in an deconvolution template. We advise to always save the deconvolution parameters next to the deconvolved image so you can easily reproduce your results and you can find it back when writing an scientific publication.
In the deconvolution Wizard you can fine-tune all deconvolution parameters one by one. The main parameter that effects the deconvolution is the acuity parameter. This can be optimized in the deconvolution setup step. If this is not sufficient you can change the stop criteria: the maximum number of iterations and the quality threshold. Selecting a different deconvolution algorithm can sometimes help as well. You can also use the deconvolution profiles/strategies as a guideline as these profiles affect the default settings of the Deconvolution Wizard. To assess the chosen deconvolution parameters you can compare the raw and deconvolved results in the Wizard or save the results and open them in the Twin Slicer or Twin MIP Renderer.
