New in Huygens 24.10: Autofluorescence and Crosstalk correction

Many biological components exhibit autofluorescence, a signal activated by excitation that differs from that of fluorescent probes. While autofluorescence can be useful for studying specific sample properties, it often poses a challenge by obscuring the signal from specific labels. Experimental protocols can help to minimize autofluorescence, such as selecting labels with excitation and emission spectra that do not overlap with the autofluorescent material. However, these measures may not entirely eliminate the issue. Autofluorescence shares similarities with crosstalk, where overlapping excitation and emission spectra from different fluorescent sources cause erroneous signal detection in the wrong channel. As such both issues can be resolved using the same tools. In this webinar, we demonstrate the new Huygens Crosstalk-Autofluorescence Correction option, which is an expansion of the previous Crosstalk Corrector. This new option can correct both crosstalk and autofluorescence in multichannel image data by looking at correlations between signal of multiple channels simultaneaously.
