Huygens Deconvolution Wizard.
Since Huygens 21.10.1p0, the Deconvolution Wizard has also undergone some major changes. As of now, the deconvolution algorithm and SNR are automatically set to ensure optimal results for each single image. Still, they can be edited according to your preference, and the Deconvolution Preview option helps you with adjusting these settings by showing a fast preview of the result within a Region Of Interest (ROI). A slider and parameter field for adjusting the 'Acuity' have been introduced to further smoothen/sharpen the deconvolution result. This allows you to leave the SNR parameter to its real SNR value.(24 min)


Huygens scripting with Python

What's New 22.10 Webinar. For the first time, Huygens Essential and Professional will have specific deconvolution options for SoRA spinning disc and for VisiTech iSIM data. The Object Analyzer now reports more information about the Region of Interest. The Look-Up Table (LUT) Editor has now an integrated Contrast Editor, and its user interface is now more user-friendly. A new Cluster Analyzer tool is available in Huygens Localizer which uses DBSCAN and FOCAL algorithm to cluster localizations in SMLM data. It also analyzes the found cluster to obtain for example the localization count or length of the clusters. For ease of use, the Cluster Analyzer contains a Tutorial which guides you through the first steps of its use. The clusters can also be analyzed as objects in the Object Analyzer. (9.46 min)

The interactive object analyzer tool for 3D microscopy images in Huygens Essential and Huygens Professional allows you to obtain statistics of individual objects by clicking on them, or analyzing all objects with a single button press. True Deconvolution

True Deconvolution Webinar The essence of using Deconvolution is to reverse the Convolution process, which is how a microscope distorts images. Since convolution involves object blurring by the microscope type-specific point spread function (PSF), it is crucial that this PSF is accurately calculated or measured, and included in a deconvolution procedure with the best possible algorithms. These are currently MLE-based as implemented by Huygens. In this webinar, we will explain image acquisition and what true deconvolution includes. Huygens Object Stabilizer


The Object Stabilizer. The Huygens Object Stabilizer can measure and correct for cell motion, thermal drift, shaking, and other types of movement (x-y-z translation and axial rotation). Both the measurement and subsequent stabilization are done in 3D and at sub-pixel level. The Stabilizer not only stabilizes 2D or 3D time series, but it also allows the alignment of z-slices within a 3D stack. (12.28 min)

Huygens Crosstalk Corrector


The Crosstalk Corrector. The Huygens Crosstalk Corrector estimates and corrects for crosstalk, also known as bleedthrough. Crosstalk can occur if there is spectral overlap between different channels, and signal is recorded from a specific dye whereas the detector is already reserved for another dye. Crosstalk in multi channel images up to 32 channels can be easily estimated, visualized, and corrected with this tool. (6.59 min)
