Getting Started

Huygens Deconvolution Wizard

Huygens Deconvolution Wizard.
Since Huygens 21.10.1p0, the Deconvolution Wizard has also undergone some major changes. As of now, the deconvolution algorithm and SNR are automatically set to ensure optimal results for each single image. Still, they can be edited according to your preference, and the Deconvolution Preview option helps you with adjusting these settings by showing a fast preview of the result within a Region Of Interest (ROI). A slider and parameter field for adjusting the 'Acuity' have been introduced to further smoothen/sharpen the deconvolution result. This allows you to leave the SNR parameter to its real SNR value.(24 min)

Huygens What's New 23.04

What's New 23.04 Webinar. Batch image processing has received much attention with the Huygens 23.04 release! The Workflow Processor contains "Automatic" deconvolution algorithm which does deconvolution with automatically found settings, as in Decon Express. The Workflow Processor now also includes a task for Crosstalk Correction and the option for performing batch Object Analysis. In addition, deconvolution has been improved for images that are acquired under sub-optimal conditions, and images and tables opened in Huygens can now be easily renamed. Huygens 23.04 also includes an update of Python from version 3.7.8 to version 3.11.1. And, via the command line of Huygens Core, it is now possible to apply a workflow template to multiple images, which makes it easy to integrate Huygens deconvolution and processing in your existing custom workflows, and to implement Huygens with a job scheduler (e.g. Slurm). Visualization & Analysis

Analyzing objects within other objects and object organization

Analyzing objects within other objects and object organization, using clusters and convex hull operations (24th of October 2023): This webinar focuses on the analysis of the organization or properties of objects, and the grouping of objects within other objects such as for example nuclei or cells. Huygens Object Analyzer can count and analyze the properties of objects within objects (cells, nucleus, organelles) in the other channel. The organization of objects can also be determined within just a single channel by clustering them using the Cluster Analyzer. And, geometric analysis of clusters is facilitated with the newly implemented convex hull option, which basically works like placing a rubber balloon around a cluster of objects. Convex hull based analysis also can aid the analysis of non-homogeneously stained objects, as is often the case with for example DAPI-stained nuclei. The ‘rubber balloon’/convex hull of the object then helps to fill the holes in the segmentation for the non-homogeneous staining. (18:20 min)

Huygens Cluster Analyzer for smlm analysis in Huygens Localizer

Huygens Cluster Analyzer (22nd of November 2022): This webinar shows you how to use the FOCAL and DBSCAN algorithm to cluster localizations in Huygens Localizer. Clusters are counted on-the-fly and rendered at the highest possible quality with ray-tracing based iso-surface rendering. You can measure volumes, localization density, width, length, and many other parameters quickly. Clear tool tips and an in-tool tutorial makes that the learning curve is really steep. (27:30 min)

(14:15 min)

Huygens Object Analyzer

Object Analysis with Huygens (3rd of July 2018): Huygens deconvolution improves resolution, contrast and signal to noise ratio in the images. Because of this, the analysis of the deconvolved images is more accurate and robust. The analysis can be performed using the Huygens Object Analyzer. The interactive object analyzer tool for 3D microscopy images in Huygens Essential and Huygens Professional allows you to obtain statistics of individual objects by clicking on them, or analyzing all objects with a single button press. The image is segmented, the objects can be filtered and grouped in Regions of Interest, and the images are then analyzed for Geometrical data, Spatial location, Relation with neighbors, Relation with reference objects, Degree of overlap (colocalization).(49.57 min)

Huygens Colocalization Analyzer

Huygens Colocalization analyzer Colocalization studies provide an important insight on several biological processes, and Huygens can help you in getting the most accurate colocalization results. Huygens deconvolution improves resolution, contrast and signal to noise ratio, reducing the effect of Blur And Noise in Colocalization studies. Our Restoration Options also correct the images for the other aberrations, getting them ready for an accurate analysis. For what concerns the analysis, the Colocalization Analyzer works more at the level of the whole image, and the Object Analyzer also provides colocalization measurements at the object level.(29.00 min)

Image Restoration

Low signal image data and Huygens

Low signal image data Webinar. Optimize image restoration and acquisition! The significant increase in signal that can be obtained with Huygens deconvolution (can be >8x) not only improves the resolution and contrast within your images, but also allows you to re-design your image acquisition experiment. By taking this approach, you can minimize photo-bleaching and toxicity issues, shorten your time sampling, and image for longer time periods. This will all greatly benefit your (time series) imaging. A special focus will be on image acquisition, super-resolution imaging, and Array (Fast-Airyscan) Detector deconvolution. (18:44 min)

True Deconvolution

True Deconvolution Webinar The essence of using Deconvolution is to reverse the Convolution process, which is how a microscope distorts images. Since convolution involves object blurring by the microscope type-specific point spread function (PSF), it is crucial that this PSF is accurately calculated or measured, and included in a deconvolution procedure with the best possible algorithms. These are currently MLE-based as implemented by Huygens. In this webinar, we will explain image acquisition and what true deconvolution includes. After this webinar, you have learned about the potential limitations of other image enhancement techniques and know how to obtain the most reliable and accurate results from your valuable image data. (27:31 min).

Huygens Stitcher

Stitching in Huygens Webinar (17th September 2019): During this webinar will discuss the Huygens Stitching & Deconvolution wizard. The Stitching & Deconvolution option is GPU accelerated, and offers a unique easy-to-use wizard for combined high-quality stitching for all types of microscopy data with the additional option to deconvolve the images, and to correct for vignetting issues. Vignetting correction can be performed manually and in a full automated fashion. The Stitching & Deconvolution Wizard is available for Huygens Essential and Huygens Professional. (14.22 min)

Huygens Crosstalk Corrector

The Crosstalk Corrector. The Huygens Crosstalk Corrector estimates and corrects for crosstalk, also known as bleedthrough. Crosstalk can occur if there is spectral overlap between different channels, and signal is recorded from a specific dye whereas the detector is already reserved for another dye. Crosstalk in multi channel images up to 32 channels can be easily estimated, visualized, and corrected with this tool. (6.59 min)
