HUYGENS Localizer: Interactive & Fast Localization Microscopy processing
The HUYGENS Localizer is a stand-alone software package for fast & high performance processing of Single Molecule Localization Microscopy data (SMLM), such as Photoactivated Localization Microscopy (PALM), Stochastic Optical Reconstruction Microscopy (STORM), Ground State Depletion (GSD), and DNA PAINT images. The Huygens Localizer is optimally designed for CPU and GPU computation, enabling a real-time processing experience. Main functionalities include:

Interactive pre-processing
Visualize the intensity flux over time, and crop useful region(s) from the total time-series. The HUYGENS Localizer is able to accurately estimate useful values from your image data, such as the photon-per-unit (ppu) value and the background. It can also estimate the PSF size from the microscopic parameters, which helps in more accurate localization results. In the particle detection stage, it is possible to interactively change settings such as threshold value to find optimal particle detection settings for your data.

Multiple CPU & GPU accelerated fitting methods
For optimal fitting, it is possible to choose from various fitting methods. All fitting methods are fully multi-threaded, and can run both on CPU-only as well as on an Nvidia GPU. The processing can be split up in block-sizes that are optimized for the (available) GPU RAM. This allows for efficient processing of tens of thousands of frames per minute.

Interactive Post-processing, Rendering & Analysis
Interactively render the localized data into a super-resolution image. Change the pixel (binning) size, and choose between various (weighted) rendering settings: bin in nearest pixel, fixed Guass, weighted Gauss, width based on uncertainty. The automatic and accurate drift correction (no fiducial markers needed!) helps to automatically remove any thermal drift for the most accurate results. With multi-channel SMLM, a shift between the channels is typically present. The Huygens Chromatic Aberration Corrector can correct for this Aberration, resulting in more reliable multi-channel image analysis. With the interactive Object Analyzer (optional), it is possible to do object based statistic analysis. Measure for example: object count, distances, volume, surface, or overlap between objects in various channels.
