This wizard driven tool guides the user through the process of distilling a Point Spread Function (PSF) from 3D bead images. It helps in selecting bead images, and in creating and saving the PSF for further deconvolution runs. Learn more.
To measure the Point Spread Function (PSF) of a microscope, one or more 3D images of Sub Resolution beads are needed. To learn more on how to record beads see: Recording Beads
Beads will only show as PSFs in the image if their diameter is sufficiently below the resolution of the used microscope. The fluorophore in the bead should preferably be identical to the fluorophore used in your samples, or at least it should absorb and emit light at similar frequencies. You can find a table with recommended beads for different types of microscopes here
