Image Quality Control webinar

Huygens Deconvolution Wizard
Huygens Deconvolution Wizard.
Since Huygens 21.10.1p0, the Deconvolution Wizard has also undergone some major changes. As of now, the deconvolution algorithm and SNR are automatically set to ensure optimal results for each single image. Still, they can be edited according to your preference, and the Deconvolution Preview option helps you with adjusting these settings by showing a fast preview of the result within a Region Of Interest (ROI). A slider and parameter field for adjusting the 'Acuity' have been introduced to further smoothen/sharpen the deconvolution result. This allows you to leave the SNR parameter to its real SNR value.(24 min)

Running pipelines via Huygens Command Line Interface, HRM, and Workflow Processor. . The Huygens Core is ideal for computer nodes, large scale 64 bit multiprocessors, and systems with multiple (high-end) Nvidia GPU cards. It can now be called from any pipeline environment using our new Command Line Interface (CLI). Run all of Huygens' Workflow Processor tasks, with a node/job managment software such as SLURM (Trademark of SCHEDMD LLC), or from an analysis pipeline scripted with for example Jupyter (Trademark of NumFOCUS foundation). Alternatively, you can use the customizable front-end Huygens HRM. We'll also address during this webinar the new tasks added to the Workflow Processor, such as crosstalk correction and the 3D Object Analyzer. (15.29 min)

Huygens What's New 24.04
What's New 24.04 Webinar. We are very excited to introduce our brand-new Image Quality Control (QC) option in this Huygens version. The QC option can be added to Huygens Essential and Professional to automatically detect the most common imaging issues, such as bleaching, clipping, hot & cold pixels, crosstalk, chromatic aberration, drift, etcetera. When detected, these issues can be further reviewed and corrected with the corresponding Huygens restoration options (see also our next webinar). Many of you will also appreciate the redesigned user interface of the Object Analyzer and Surface Renderer, which has become much more user-friendly with extendable panels, various new icons for easy navigation, a bar for your favorite buttons, and much more. Within the Object Tracker, it is now easier to detect and fix possible flaws in the tracks and to add missed nodes to tracks. In addition to these new features, the Batch Feeder allows you now to change the depth of subfolders of the input data, to recursively search for images, and to preserve the folder structure of subfolders as found in the input folder in the output folder. Huygens STED deconvolution and optimal processing of STED data (March 23th, 2023): Whether you are performing STED imaging on an Abberior, Leica or custom-build microscope, optimal STED deconvolution requires a PSF that considers specific STED parameters such as saturation and immunity of the fluorophores, and percentage of 3D depletion. Analyzing objects within other objects and object organization
Analyzing objects within other objects and object organization, using clusters and convex hull operations (24th of October 2023): This webinar focuses on the analysis of the organization or properties of objects, and the grouping of objects within other objects such as for example nuclei or cells. Huygens Object Analyzer can count and analyze the properties of objects within objects (cells, nucleus, organelles) in the other channel. The organization of objects can also be determined within just a single channel by clustering them using the Cluster Analyzer. And, geometric analysis of clusters is facilitated with the newly implemented convex hull option, which basically works like placing a rubber balloon around a cluster of objects. Convex hull based analysis also can aid the analysis of non-homogeneously stained objects, as is often the case with for example DAPI-stained nuclei. The ‘rubber balloon’/convex hull of the object then helps to fill the holes in the segmentation for the non-homogeneous staining. (18:20 min)

Huygens deconvolution improves resolution, contrast and signal to noise ratio, reducing the effect of Blur And Noise in Colocalization studies. Our Restoration Options also correct the images for the other aberrations, getting them ready for an accurate analysis. Low signal image data and Huygens

The significant increase in signal that can be obtained with Huygens deconvolution (can be >8x) not only improves the resolution and contrast within your images, but also allows you to re-design your image acquisition experiment. True Deconvolution
True Deconvolution Webinar The essence of using Deconvolution is to reverse the Convolution process, which is how a microscope distorts images. Since convolution involves object blurring by the microscope type-specific point spread function (PSF), it is crucial that this PSF is accurately calculated or measured, and included in a deconvolution procedure with the best possible algorithms. Huygens Object Stabilizer
The Object Stabilizer. The Huygens Object Stabilizer can measure and correct for cell motion, thermal drift, shaking, and other types of movement (x-y-z translation and axial rotation). Both the measurement and subsequent stabilization are done in 3D and at sub-pixel level. Huygens Crosstalk Corrector
The Crosstalk Corrector. The Huygens Crosstalk Corrector estimates and corrects for crosstalk, also known as bleedthrough. Crosstalk can occur if there is spectral overlap between different channels, and signal is recorded from a specific dye whereas the detector is already reserved for another dye. Crosstalk in multi channel images up to 32 channels can be easily estimated, visualized, and corrected with this tool. (6.59 min)
