HUYGENS Localizer: 快速互动处理

Localizer 是一个包含在惠更斯下载包中但能独立使用的软件,支持单分子分辨率定位显微镜系统,支持 Photoactivated Localization Microscopy (PALM), Stochastic Optical Reconstruction Microscopy (STORM), Ground State Depletion (GSD), and DNA PAINT 数据,支持 CPU 与 GPU 显卡并用,并具以下功能:

Localizer 重构前处理

为单分子分辨率定位数据作出重构前的处理,例如:对时间序列信号 intensity flux 或图像的剪裁,对采集信号的相机的数据 photon-per-unit (ppu) 输入与设定、背景的估计...等,以上数据使重构结果更准确。 定位过程 particle detection 中,用户亦能透过即时预览与互动介面设定定位的敏感度。

The pre-processing steps and particle identification settings can be adjusted with real-time visual feedback on the identified particles. With the time-slider you can also quickly obtain feedback on the settings for different time-points.

重构后分析、渲染与输出适合发表用结果图像

渲染功能具备多个不同设定,例如: 像素 (合并 binning) 大小,各种(加权)渲染设置:最近像素中的合并 (bin in nearest pixel ) ,固定高斯 (fixed Gauss),加权高斯 (weighted Gauss),基于不确定性 uncertainty 的宽度 width based on uncertainty;而且用户亦能选择位移校正(不需特别校准标签);对于多通道图像,惠更斯 Localizer 提供进一步的色差校正;分析功能也包含在 Localizer 中, 例如:对象数量,距离,体积,表面或各个通道中对象之间的重叠。

HUYGENS Localizer Image Gallery

Nanorulers Cac
GATTA-PAINT 80RG (2x 80nm) nanorulers imaged with STORM system. Both the widefield (accumulated) data (background subtracted), and the processed image show a clear shift between the two channels. This can be accurately corrected with the HUYGENS Chromatic Aberration Corrector. The determined Chromatic Aberration template can be used for correcting chromatic aberration for other images as well. Image data provided by GATTAquant GmbH, and processed with HUYGENS Localizer.
