Huygens 软件更新消息

惠更斯 2017 年春季版 17.04 已经推出了( 2017 年 5 月 ) !

摘要

Light Sheet/ SPIM Fusion Wizard 光片照片的 Fusion 与去卷积 Wizard 功能,这个功能对 MultipleView SPIM 的数据;或激发光片固定在同位置,但样本以不同角度旋转采集得的图像均可以使用。惠更斯 Huygens 会把原始数据作去卷积处理再以 Fusion 组合数据,光片照明的散射亦同时得到校正。

支持多张显卡 GPU 的批量处理 Batch Processor 加速

批量处理 Batch Processor 加入更多对 Object Stabilizer 的选项,自由度更大,更弹性。

支持高清 4K 屏幕

惠更斯的介面提供亮度低的暗色皮肤作选择,黑暗的实验室采图与图像处理时,提供对视觉较舒适的介面,当然惠更斯原来的皮肤颜色仍是选项之一。

光漂白校正 Bleaching Corrector,对 XYZ , XYT , XYZT 作用测量与校正。

只需一个按键 Deconvolve 就自动快速处理,快速处理的条件将由惠更斯 Huygens 作自动估计,测量,图像的讯噪比与所需数据 metadate 会由惠更斯 Huygens 以图像的讯号与结构作出智能评估再直接进行去卷积处理,方便快捷但以结果准确计算作优先条件!

Huygens Titan

  • 当 Titan 读取庞大的 *.LIF 档时,Titan 早在索引 (indexing) 的时候,把 *.LIF 文件结构中的各图像及资料读取。 *.LIF 文件中含多图像,Titan 也能分辨出来。
  • 为了减少电脑处理器的负担并增加工作效率,当 Titan 遇上庞大的文件档,Titan 利用 placeholder 显示出 thumbnail 并处理其他的图像搜索,使 indexing 继续进行。
  • Titan 搜索能力的加强,文件中的图像是以时间序列的图像或一文件中含有多图像的档案都可以分辨。

Huygens Professional

  • Stitcher 拼图功能
    • 加入了暗角校正
    • 增加了工作效率及更有效的利用记忆体。
    • 能教正原始 *.LIF 档的每一块图像的方向。

Huygens Essential

  • 加入了与 Leica LAS X 的互动,图像可从 Huygens 直接切换到 LAS AF X 或从 LAS AF X 切换回 Essential 。

Bug Fixes

(All) In license mechanism: string licenses with no extra flags did not get recognized.
(All) In LSM file reader: Emission wavelength was not estimated accurately for low pass and short pass filters.
(E/P/C) In Batch processor and HuCore jobs: the sampling distances were not properly loaded from the templates, instead the metadata values were used.
(P/S/C) In ICS file writer: data compression was not enabled as intended.
(E/P) In Batch Processor: the selected sub image did not get processed. Instead, a different sub image from the same data set was deconvolved.

全新的方法去处理 PSF 形状的改变
点扩散函数 PSF 常常受到介质的改变而导致球面色差的影响,结果导致 PSF 形状的改变,这个问题常随着样本的深度而产生。之前的解决方法是把图像分割成小块,每一小块具备有本身的 PSF ,这是一个有效率的方法,可是也会做成失真的效果。新的方法叫 "sub-brick" 方法,把基于 Fast Fourier Transform (FFT) 卷积操作成为去卷积计算的核心换成多个 FFTs ,而每一个具备不同的 PSFs,这个新的计算方法可使用于 CMLE, QLME 及新的 GLME 去卷积模式中。

GLME 全名为 Good Roughness Likihood Estimation,这个模式是基于最大后验概率(Maximum a Posteriori probability) 来进行计算,这个模式正适合使用于 PSF 形状的改变。
GMLE 是处理STED 数据的预设模式,其他系统进行去卷积时需要到 Expert Mode 去设定 GMLE 模式。

  • 您可以为您的图像加上文字记号或注解,这个记号只用在 Titan 中,并不改变您的文件的档案,而文件具备同一个记号可以整理成为一个项目 (Project)。
