Both Abberior's STEDYCON and FACILITY line microscopes include an efficient connection to the Huygens Software. This feature allows researchers get the most out of their high-quality STED images by using the powerful and renowned Huygens deconvolution MLE algorithms and image analysis modules in a stand-alone Huygens Professional package.

Why HUYGENS? Get more from your data!

STED is an intrinsic super-resolution technique (i.e. no post-processing is required to obtain super-resolution) and can easily reach a resolution below 40 nm. However, even with highly sensitive photon-counting detectors, the physics behind STED imaging and the intrinsic photon shot-noise, limit the photon counts per pixel which leads to noisier images compared to the confocal counter-part. This can be partially solved by increasing line-accumulation, i.e. scanning the same lines multiple times, but this has the downside to slow down the image acquisition, and tends to result in more photo-bleaching issues and may cause cell or sample damage. This is where Huygens will help.

The Huygens Professional package offered with the STEDYCON and FACILITY Line (see below for more details), helps researchers to get more from their confocal and super-resolution STED images, even with low signal and with noisy images. The Huygens CMLE and GMLE deconvolution algorithms have shown to perform extremely well on dim and very noisy STED data, enabling users to image much faster and with less line-accumulation. In turn this allows users to acquire images much faster, reduce bleaching and minimize cell damage. The powerful automatic and/or manual restoration options in Huygens make the images more reliable and easier to visualize and analyze, enabling researchers to save a significant amount of time with their imaging and image processing workflow.

Connection to HUYGENS Professional a optimized workflow

Huygens now fully supports the OBF data format used for the STEDYCON and FACILITY line. Users can open OBF data directly in Huygens via drag-and-drop or via the regular open command. Huygens automatically reads in all meta-data required for accurate deconvolution and further processing.* The acquisition software of Abberior also introduces a feature that allows users to send any image from their imaging session to Huygens Professional with a single mouse-click. The dataset can be easily deconvolved in Huygens via the fully automated Deconvolution Express, or with the Deconvolution Wizard when the users prefers full manual control on the deconvolution settings. In addition, also the Abberior's MSR files, that is used for their other STED microscopes can be read into Huygens.

* For optimal support of OBF meta-data in Huygens, the data should be acquired with version 4.0 of the STEDYCON software, and Huygens version 19.04 or higher is recommended. Data acquired with version 6.0 of the STEDYCON or with the Abberior Facility Line requires Huygens versions 21.04 or higher. A license flag (I) is required in the Huygens license in order to make use of the Abberior Image Feeder functionality. Contact us if you require a test license.

HUYGENS for Abberior: what's included?

Abberior Instruments offers various Huygens Professional configurations, optimized for their STED microscopes and tailored to fit your various image analysis needs. A powerful, yet very affordable and compact GPU acceleration workstation can be optionally added to your configuration for most efficient image processing. The workstation specs have been selected by SVI and Abberior Instruments specialists to ensure an optimal supported solution.

The HUYGENS for Abberior configurations include a stand-alone Huygens Professional license and includes many useful functionalities such as: Deconvolution Wizard, Deconvolution Express, Workflow Processor and various easy-to-use 2D and 3D visualization tools. The table below provides an overview of the various Huygens packages offered by Abberior Instruments.

It is always possible to extend your Huygens license with additional options. Any Huygens package can be installed on the acquisition system, but it is also possible to install/migrate the Huygens license to a separate workstation or upgrade the license to an floating Huygens Everywhere license that can be used on any computer.
